研究领域

针对神经科学的重大前沿问题---记忆的获取、储存和提取机制开展研究,涉及宏观层面行为学药理学、介观层面神经环路和微观层面的细胞分子生物学技术方法。同时,针对相关脑重大疾病开展新药和新技术发现、药理药效学等研发,为这些脑疾病的诊断、治疗提供新途径。


1、海马环路与突触可塑性---基础研究

四十多年来,国际学习记忆研究领域取得了丰硕的卓越成果,例如发现和证明了突触传递效率长时程增强(LTP)和减弱(LTD)是学习记忆的重要细胞分子机理。尽管如此,学习记忆研究领域仍然面临着巨大挑战:还几乎完全不清楚记忆以什么形式储存在哪里、记忆又怎么被提取等。由于海马在记忆形成和提取中起着重要作用,我们试图探索海马各亚区如何处理信息、又如何与其它脑区进行信息交流,最终实现记忆的储存和提取。拟采用多通路突触传递记录、微电极阵列神经放电记录、光遗传、基因操作和行为药理等技术来研究这些重大的科学问题。

2、基于异常记忆的动物模型与精神疾病---应用基础研究

大脑无时无刻处理着海量信息,然而仅有少量信息以未知方式被编码并储存为长时记忆,绝大多数信息被迅速遗忘。记忆和遗忘这一对矛盾遵循着自然界“动态平衡”法则。一旦这一平衡法则被打破,既可导致“忘不了”如创伤后应激综合症(PTSD)、抑郁症和成瘾,也可导致“记不住”如孤独症和老年痴呆症(AD)。这些异常记忆相关的脑疾病均存在严重的精神障碍或残疾,已导致了沉重的社会和经济负担,其发病机理还不完全清楚,仍然缺乏有效的临床预防、诊断和治疗措施。我们试图建立异常的记忆和遗忘动物模型,研究其细胞分子机理,进而阐述为什么一些情况下如急性应激相关的记忆就难以遗忘,而另一些情况下如慢性应激相关的记忆容易遗忘甚至过度遗忘。重要的是通过人类疾病的动物模型不仅可找到诊断和治疗新措施,也可观测疾病的发生发展过程,进而可能找到有效的早期预防和干预措施。这些应用基础研究进展又为深入理解记忆的储存和提取机理等重大基础科学问题提供独特的视角。

3、抗精神疾病新药的药理药效学---应用研究

神经系统区别于其它系统的最显著特点在于神经细胞间通过上千个突触进行信息传递构成功能性神经网络,并且突触信息传递效率具有普遍的高度可塑性如LTP和LTD。重要的是,从昆虫、鸟类、哺乳类到人类,突触结构和分子构成高度保守,其传递效率的可塑性也高度保守,充分展现了系统演化过程中在突触水平上动物与人类的高度相似性。然而随着神经细胞数量急剧增加、以及神经细胞构成的神经环路和调控机理的复杂性高度增加,从动物到人类也存在巨大的、难以跨越的差异性:更卓越的智慧、更高级的认知能力和更复杂的行为。神经系统新药的研发就面临着这样的一致性和差异性挑战。因此,神经系统新药的药理药效学研究必然涉及两大系统检测:证明新药在人类疾病的动物模型中有效;证明新药在突触传递和突触可塑性中也有效以及新药的作用靶点和机理。我们已经建立了实现上述检测系统的技术手段,成功地完成了抗AD(酚克络酮)、抗抑郁症(奥生乐赛特)两个一类新药的全套药理药效学研究,目前正在II期临床试验中。还可针对孤独症、焦虑症、抑郁症、精神分裂症、毒品成瘾、慢性疼痛、PTSD、AD、缺血性中风等脑疾病,系统研究治疗新药的药效学和药理学。我们正在针对AD、PTSD、血性中风后神经保护开展中药来源的活性物质筛选和新药研发。
