Research Areas

Professor Nai-Xing Wang, a well-known organic synthetic chemist, was validated as an academician of the Romanian Academy of Technical Sciences in 2022. He serves as a professor and PhD student supervisor at the Institute of Physics and Chemistry of the Chinese Academy of Sciences from 2000 to now. In 1996, he went to the United States for postdoctoral research and received the Robert A. Welch Postdoctoral Fellowship in Rice university in 1998. In 2000, he returned to China and was selected as the "Hundred Talents Program" of the Chinese Academy of Sciences. He has published multiple papers in core Journals of organic chemistry, including Org. Lett.; J. Org. Chem.; Adv. Synth. Catal.; Coordin. Chem. Rev. and Nature, with a total of over 180 papers. A paper published in Eur. J. Org. Chem. (2017, 5821–5851), has been cited 428 times; Org. Lett. 2008, 10, 1179 (cited 92 times by others); Org. Lett. 2008, 10, 1875 (cited 86 times by others); Org. Lett. 2015, 17, 4460 (cited 81 times by others); Org. Lett. 2016, 18, 5986 (cited 63 times by others); Coordin. Chem. Rev. 2012, 256, 938 (cited 72 times by others). He has been granted 12 invention patents. In 2013, he was awarded the Science and Technology Award of the Beijing city government for developing multi-chiral novel coenzyme NAD(P)H model molecules and the synthesis and transfer development of multi-chiral drug Nebivolol.

Professor Nai-Xing Wang's research achievements in synthetic organic chemistry over the past decades have had a significant impact on synthetic organic chemistry, which primarily focused on:

1)Developing the Wang reaction;

2) Synthesizing and studying the properties of novel NADH coenzyme model molecules;

3) Synthesizing chiral drug molecules.

1. Research on new C(sp3)-H bond difunctionalization reactions of styrene derivatives with solvent molecules (Wang reaction)

Professor Nai-Xing Wang has made a series of progress in the C(sp3)-H bond difunctionalization reactions of styrene with alcohols, ketones, nitriles, ethers and thioethers (Wang reaction), which is carried out through a free radical relay process. A lot of articles have been published in core organic chemistry journals, these work have been reviewed and summarized into a Wang's reaction formula (Wang's reaction) in Synthesis (Synthesis 2019,51,4542). Professor Nai-Xing Wang was invited to publish an account article on Synlett (Synlett 2021, 32, 23), systematically summarizing the difunctional reactions of related styrene derivatives with solvent molecules as a Wang reaction by a general formula. In June 2026, Liu et al. provided a detailed and relatively comprehensive review of the main contents of the Wang reaction in an extensive article in RSC Adv. 2026, 16, 31348 (IF 6.1).

Wang reaction could can be summarized as:

Wang summarized the universal reaction mechanism formula of the Wang’s reaction based on the experimental results:

General formula of reaction mechanism for Wang reaction.

2 Synthesis and characteristic study of novel NADH coenzyme model molecules

Wang and co-workers has designed and synthesized the first C3 symmetric novel chiral NADH(Nicotinamide Adenine Diunclotide Hydrogen) model molecule with a special bowl-shaped conformation, this novel C3 symmetric NADH model is capable of fluorescent emission at 455 nm when excited at 390 nm. (Wang: Adv. Synth. Catal. 2009, 351, 3045.)

In 2015, Wang and co-workers designed and synthesized a petal-type chiral NADH model molecule, and researched on the asymmetric reduction and fluorogenic sensing for Fe3+. The research results could provide a new strategy for the design of various NADH models based fluorescent chemosensors.(Wang: Sci. Rep. 2015, 5, 17458-17464.)

Wang's research on NADH completely breaks through the limitations of previous researches in this field, in the past, chemists only used NAD(P)H model molecules for organic reduction, Nai-Xing Wang pioneers a new method for efficiently reducing stable unsaturated bonds using dehydrogenases to catalyze NAD(P)H model molecules. Wang was invited to publish Account articles on NAD(P)H (Synlett 2007, 18, 2785 and Synlett 2017, 28, 402).

3 Synthesis of multi chiral antihypertensive drug with specific effect: Nebivolol and Acebutolol

3.1 Synthesis of Nebivolol

In 2007, Nai-Xing Wang and co-workers synthesized multi chiral specific effect antihypertensive drug Nebivolol with four chiral centers. The traditional method of constructing stereocenters using noble metal complexes as chiral catalysts, which is difficult to industrialize, A new method utilizing natural chiral source mannitol has been developed to synthesize the highly effective antihypertensive drug Nebivolol by Wang, The strategy ingeniously utilizes mannitol to obtain the chiral synthons R-acetone glyceraldehyde acetal. After condensation and cyclization, two intermediate configurations, (S, R) and (R, R), are obtained. The intermediates are bonded to yield the absolute configuration of the stereospecific target product, S,R,R,R-stereoisomer (see Wang’s work in: Synlett 2005, 9, 1465 and Synthesis 2007, 8, 1154). Wang’s strategies can easily construct multiple chiral absolute stereoisomers. The final research results were successfully transferred to Beijing Maijin Biopharmaceutical Company in 2015 and was industrialized (Nai-Xing Wang was awarded the Science and Technology Prize of the People's Government of Beijing Municipality).

The synthetic route of multi chiral specific effect antihypertensive drug Nebivolol is as follows: (Wang: Synthesis 2007, 8, 1154)

3.2 Synthesis of Acebutolol

In 2009, Wang and co-workers synthesized a chiral high-efficiency antihypertensive drug with High Enantioselectivity: Acebutolol, (R)-N-[3-Acetyl-4- (2-Hydroxy-3-Isopropylamino- Propoxy)Phenyl]- Butanamide. Synthetic strategy for chiral specific effect antihypertensive drug Acebutolol is as follows: (Wang: Sci. China Chem. 2009, 52, 1216.)

Nai-Xing Wang's group has made some contributions in the field of sulfur-containing new reagents from 2015 to 2021, four sulfur-containing new reagents were developed. DMSO-I2 (Adv. Synth. Catal. 2019, 361, 3008.) and DMSO-TsCl (Eur. J. Org. Chem. 2021, 1446.) are excellent sulfur methylation reagents. Na2S-CH3CN reagent is a cascade reaction reagent for generating sulfoxides (Curr. Org. Synth. 2016, 13, 651.). Especially the new sulfur-containing reagent Na2S2O4-TBHP (Sci. Rep. 2016, 6, 20163.) discovered by Wang's group on February 2016, it was found that aromatic alcohols can be selectively oxidized to aldehydes by Na2S2O4-TBHP, and Wang's group also researched on the reaction mechanism. In April 14, 2016, China Science Daily reported this discovery. Na2S2O4-TBHP reagent was called as the Wang’s reagent by JECI, 2023, 8, 269, and in 2024, Synth Comm. reviewed on the Wang’s reagent (Synth Comm. 2024, 54, 1785.), and Science and Technology Review (2024, 42, 98.) in China also introduced this Wang’s reagent.

Research Interests

    Professor Wang completed the total synthesis of one complicated biologically and pharmacologically active molecule (Nebivolol) with four chiral centers by developing natural-chirality-origin asymmetric synthesis. Some very skillful method and strategies were used and some expensive reagents were avoided. Some theoretic problems were solved also through the calculation of quantum chemistry. ( Synthesis 2007, 8, 1154.; Synlett 2005, 9, 1465.)
    A novel chiral NADH model with C3 symmetry was synthesized with a view to developing new chemoselective and enantioselective reducing agents by Prof. Nai-Xing Wang. Its special bowl-shaped conformation could encase and fix some substrates and three hydrogens at the C4 position of all dihydropyridine rings in the inner part of the bowl could transfer to the substrate, a novel powerful reducing agent was obtained, an account paper was published in Synlett in 2007, and research paper about it was published in 2009. ( Adv. Synth. Catal. 2009, 351, 3045.)
    Professor Nai-Xing Wang research on the new methodology of organic synthesis, especially on the asymmetric synthesis. A lot of good paper were published on the core journals of organic chemistry, such as Org. lett.; J. Org. Chem.; Adv. Synth. Catal.; Coordin. Chem. Rev. and Nature. So far, more than 180 papers have been published in scientific journals in both foreign and domestic such as Sci. China Chem. And the most recent one is Eur. J. Org. Chem. 2017, 5821, which has 408 citations; Org. Lett. 2008, 10, 1179 (citations: 92);Org. Lett. 2008, 10, 1875 (citations: 86);Org. Lett. 2015, 17, 4460 (citations: 73);Org. Lett. 2016, 18, 5986 (citations: 53); Coordin. Chem. Rev. 2012, 256, 938 (citations: 72). Professor Wang obtained 11 Authorized  patents and published four monographs, and also served as a reviewer for core professional publications of ACS in the United States.

     Professor Wang once published many papers about the synthesis of polynitrogen derivatives of Fullerene and published some papers in this field, such as J. Phys. Chem.; Carbon; Tetrahedron and Chem. Phys. Lett. and so on. Before in 1992, professor Wang once researched on the synthesis of some heterocyclic systems benzofuroxan compounds which could be used as energetic materials, more than ten papers were published on this field, such as in PEP Journal of Wiley-VCH.
